HIV Research Breakthrough Brings Cure Closer

Scientists finally crack the structure of the enzyme integrase, which HIV uses to infect host cells

UK experts at the Imperial College London (ICL) have recently made a significant breakthrough in HIV research, after more than 20 years of research. The achievement could have considerable implications on approaching HIV and AIDS patients, and could result in groundbreaking, new therapies for the condition, healthcare experts say. Details of the investigation appear in the latest issue of the respected scientific journal Nature, e! Science News reports.

Being a virus, HIV cannot replicate on its own, and needs a host cell in order to produce more copies of itself. So, when it enters a human cell, for example, it hijacks the genetic mechanisms at work inside, and makes them produce more viral agents. It therefore needs to paste a copy of its genetic material into the genome of the cell, and the way it does that is via an enzyme called integrase. What the ICL team did was grow a crystal that revealed the structure of integrase. This could help molecular biologists create drugs that are targeted at this enzyme directly, a completely new approach to treating HIV.

“It is a truly amazing story. When we started out, we knew that the project was very difficult, and that many tricks had already been tried and given up by others long ago. Therefore, we went back to square one and started by looking for a better model of HIV integrase, which could be more amenable for crystallization. Despite initially painstakingly slow progress and very many failed attempts, we did not give up and our effort was finally rewarded,” ICL Department of Medicine expert Dr. Peter Cherepanov says. He has also been the lead author of the Nature paper.

The investigation was conducted over a period of four years, during which time more than 40,000 trials were attempted. Of these trials, just seven resulted in usable crystals. These materials are absolutely essential for studying molecules. The enzyme was collected from a type of retrovirus known as the Prototype Foamy Virus (PFV). In this organism, the integration processes and functions of integrase are roughly the same as the ones in HPV, the researchers say. They add that the final data showed that the enzyme had a very different structure than the one proposed through the theoretical model. The new data will soon help inform on new drug-design efforts.

The article mistakenly states, "This could help molecular biologists create drugs that are targeted at this enzyme directly, a completely new approach to treating HIV." In fact integrase inhibitors already exist, one is FDA-approved and performing very well (ralegravir/Isentress). Scientists were able to come up with this compound despite not fully knowing it's mechanism of action. This new discovery showing the structure of the integrase enzyme may one day lead to better integrase inhibiting compounds, maybe some that the virus will not be able to evade through resistance.

I hate to break it to you, but there's no guarantee that this will really lead to a cure, and even if it does, it could be decades before it's widely available. Such potential cures have always had to go through animal trials for years before they could be tested on any human subjects. The absolute fastest I can imagine a cure coming from this is ten years, and that's if it gets done in a big hurry.

@bobber You can humanise the immune systems of many animals in a lab environment. This is done quite regularly with mice. I has already been shown that zinc finger technology can be used to disable the ccr5-d32 gene in lymphocytes which thencan be infused back into the patient, providing profound 'control' over the virus.. doing away with the need for antiretroviral drugs. it is not a sterilising cure, but it is certainly a 'functional cure'.. this is the way the world will move if you ask me..

I consider this to be good news and I believe that there is a cure for HIV; first and foremost, one must have FAITH in the Creator who can and will remove all; Second, one must be obedient to his WILL to see that all things are remove in HIS time and not in thy own; doing what is required is mandatory, eating healthy, sleeping and feeding the soul on daily basis will heal anything while taking extra steps to enhance them;

A total cure for HIV wont be readily available for several years. Testing and funding are things that take years to gain. However, HIV and AIDS are not the death sentences they used to be. Anti-retrovirals can bring the virus to a stand-still and even push it into hybernation-like state if caught early enough. People with AIDS can still live well into old age without major complications. A lot of it has to do with taking care of your body. Remember, HIV attacks the immune system. KEEP YOUR BODY HEALTHY! The human body is truly amazing, take care of it and even without anti-retrovirals you can live a long and happy life with the virus in you. Trust me, I've had the virus for 8 years and my doctor says my blood and immune system look healthier than his. If I can live through this, so can you.

assad i lived with hiv for 10 years with out meds and lived a full positive life.i was a tv actor.the last 8 yrs((im hiv 18yrs)i have osteporosis becuase of the meds.also i have had lipodystropy for the the last 7yrs which has changes my appearance dramaticaly and my quality to my life has declind to the point of where i am a shadow to my self.dont preach to people my friend.i wish you good health but think before you speak and where your words land.love and light to you damien

well this looks very promising, im sure most of the researchers are true in their efforts but funding is the major issue, with todays technology and what we know today if there was funding im sure a cure or even a way to make the virus permanently dormant would be possible, but we should keep in mind that big pharmacuticals will do anything they can to hide this as they make millions from selling arvs. its sad how human nature is and how leaders of the world cannot do much on such important issues but never loose hope

Research is fine for the cure, what about vaccine first which can prevent from happening , ie hiv virus needs a host cell to make a copy, why can't they develop a vaccine to prevent this.
Lets hope there will be cure in year or 2.

Lets hope there is cure in the nearer future... Not nice wondering when my times is up. I get scared when i have 1 sip of beer or miss a meal.
Trying to be healthy is HARD work.

Doctors i have spoken to have said that the cure is long long away... I argued that if the ARVs suppress the virus then why couldn't they killed it?? They said its because there's hidden virus in the brain or that the ARVs is not strong enough to reach all place...

I pray every day and check online now and again in the hope that the headline would say "CURE BREAKTHROUGH" only to find that they have discovered how they work but nowhere near cure if not that close... But i am very hopeful that there will be cure... but its just the WHEN is what bugs me... I hope its just round the corner... Why can't all the sciencist in the world who work on it to work together! who cares about who found this method or that method first.. money is not important when we have our health to worry about and the future generation.. if there is no cure soon it will be disaster for our future generation. I got it through snorting coke (sharing straw) not realising you can catch it through that although the risks is very small.. but if i didn't know this then surely there's more ways to catch it that people wouldn't even know... what does that say...
Congrat to those devote science people who still work on it.. Lets do our bit by involving charities and rise money for the researchers and sciencists so that they can continue with the work.

I don't think we'll see a cure anytime soon. Pharmaceutical companies make too much money keeping HIV patients on their toxic drugs. Once in a while their research team make the headlines with a pseudo discovery that will bring hope of a cure and bring more funds for research. They just care about money...

I hope there is cure in Ghana a man Dr Akuamoah Boateng who is a doctor have joined heads with some scientist in the US to find a herbal cure but people don't believe it,the herbal medicine is called mab formula 1 and mab formula 2 which kills the virus completely and boots up the immune system in advance.

Hi we thank all people, consultant, Doctors who are involved in find the hiv-aids cure. The problems is that all drugs have side effect especiall the lipodostropy. We just now need God's intervetion to completely take control and remove this curse from us. The drugs are just to keep us on and on as they damage the system. Do not be desparate. Of course its * to find out you are infected with HIV, but life does not end at that same moment. Hold your self together and make sure you are there when the cure gets to us. I'm also 17 years old and infected. The truth is you can have a family, and you can find love. You're not going to die tomorow unexpectedly. It's not really a big deal, you use a condom and thats it. You have 30 years in front of you. Who knows how many people get hit by a car every day!

Bad things happen all the time, but that's life. Life isn't pristine health till you're old and suddenly something comes up. Stay optimistic, life goes on.

hope when they find a cure it becomes available to everyone and doesn't get swept under the carpet in order for the pharmacutical companies to continue making billions, as for the seventeen year old, keep it together and be strong you can have your own family if you are a female it very easy and there is medication to prevent your unborn child from being born positive and if you are a male there is sperm washing so you won't infect your partner. It will be the greatest news ever all world over.We can now start to imagine a generation free from HIV/AIDS.Though there's ARVs to prolong life for those with hiv, its really a drug burden to take medication everyday for life. We urgently need a cure. just imagine the kids born with the virus, its really miserable to see them take medication day in day out.even with the ARVs, these children dont live an absolutely free and happy lives.Doctors, researchers, physicians etc, please work so hard around the clock, a cure is urgently needed.

thank you all for the positive messages, us with the virus sometimes completely loose hope even with ARVs because of the stigima and toxicity of the drugs.Docs say they damage the liver, kidneys after a long time of use.WE NEED A CURE PLEASE.

I am 23 and HIV I admire you guys living with the virus for so many years and still having faith and hope. I on the other hand feel like I died already. I think about my death every singel day and I am longin for it. If there is a God out there, he has forgotten about me.

you will survive and get to be amongst the first who are cured. There are reasons HIV is important. Science is finding this virus to be like the Space Program. It is leading to deep and far-reaching understanding of disease, genetics and a lot more. They have a functional cure already which takes your own cells out and alters them genetically and replaces them later in a few months. That is only one way. The new mosaic vaccine, funded by Gates, is using a computer to generate more mutations than the virus can make, and programs it into the vaccine. This goes into human testing in 2012. It already shows a VERY strong immune response in animals. A third way is a new particle that infects the HIV virus itself. It cannot attach to anything else, so is harmless. It destroys the genes in the HIV, so that when the HIV multiplies it does not make more HIV, but more of these PARTICLES, which infect more of the HIV. All 3 of these methods will work, but the vaccine is the easiest/cheapest. You have good reason to expect to be cured, and this is real. Don't go into denial and take risks, or abandon your meds. Do what you can to clean up the body from the med-toxins..but for now the meds are what keeps you going, nothing else will. Drinking water, exercising, eat well as you can afford. And there will come a day when it is realized that everyone with HIV is in fact a hero, or heroine!

bG, can you provide a link or more information this the "new particle that infects the HIV". If this is to serve as a cure, I am very hopeful. The science behind this seems sound. However, i cannot find any information on this. As long as there is something to hold on to, i have hope, without information like this, life seems rather empty. I feel as though i can now experience a modicum of happiness as long as this information is true. I cannot handle having my hopes crushed. I will not survive it.

My opinion on HIV is basic; To cure HIV find the source. I think I know the source and it is not from monkeys because if it was the question still stands, where did the monkeys get the SIV from? Hi there, I just want to first say stress will be your biggest enemy. I have talked to several people who found out when their CD4 was at 50 and below... now they are on meds and up past the 300's and one i know is at 1100 already. its terrifying at first, i know, i am recently diagnosed... but your best bet is to talk to people who are doing awesome. i go on hivnet.com to the chat room and talk to people and they are great at support. Youll be fine. the only thing i can say is listen to your dr's. dont skimp and just go to whoever... go to the best you can find. also, most of my drs believe that their will be a cure in 10 years but no matter what we will always test positive for the virus. its no different than mono, chicken pox, measles etc... if you were to take those tests you will still test positive because the antibodies are still present in your system. also, its great that you believe in god, but HE will not cure you. you need to be dependent on your medical team and follow their instructions. that being said... take care and dont worry about it... youll be fine

Is there any more news on this? It's been a long time since the article. Has anyone noticed how a new AIDS/HIV breakthrough seems to come out every so often and then we hear nothing more about it... its as if it's all hype? For example, is the treatment available at the doctor yet? Can we see a schedule of planned testing phases, and get updates etc.? Its as if they report it and it always vanishes into the ether.

in zambia herbs are also on research but they is need to look at them very well in 2006 clinical trial were undertaken on 26 HIV postive people there only thing that showed is that herbs can prolonge life as well due to lack of funds the trails were onnly for 6 months but the results were very good to these who undertook the trails . it is very imporatat that all country come together and find and support each other in the research for the cure we vshould share our findings from the baseline look at the CD4 VRIOLAOD and other we are man and weman who can make a charge

thanx 4 the gud nuz. i feel when such gud nuz is around the corner, such reputable scientists/researchers shudnt let it jus go lyk that. they shud take pararell progressions of their experiments with some giving hourly results, daily results, weekly results, monthly, yearly and some 5 year results continuously and simulteneously. i believe with such force and proper funding best nuz is coming lyk tomorow. hey please assist the needy. thank u.
